Preparation of novel polymeric monolith and its application in enzymatic kinetics study

Polymeric monolith materials have received much interest due to their high rates of mass transfer and large surface area. In recent years, the preparation of the polymer monoliths was carried out and the protease were efficiently immobilized on the materials by Qi’s group. The novel enzymatic reactors were applied for the hydrolysis of proteins (Fig. 1). It has been found that the enzyme efficiency could increase 300 times comparing with the free enzymatic solution (Journal of Materials Chemistry, 2009, 19, 767-772;Analytica Chimica Acta, 2011, 692, 131-137;Journal of Materials Chemistry B, 2013, 1, 2260-2267).

Then the monoliths were polymerized in capillary and used for the amino acids enzyme immobilization. Based on the chiral separation of D,L-amino acids by CE/MCE, the methods of kinetics study of amino acids enzyme were established (Fig. 2). Moreover, the inhibitors screening of amino acids enzyme was investigated. Further, combination with the dialysis system in living mice, the change of amino acid enzyme activity with the kidney ischemia was explored (Analytica Chimica Acta, 2011, 691, 103-109; Amino Acids, 2012, 42, 337-342; Electrophoresis, 2013, 34, 409-416). These methods have provided new tools which could be applied in understanding the chemical essence involved in some physiological and pathological events.
